She studied and graduated from Hewlett High School. Afterwards, went on to The New School university's famous art college, Parsons The New School for Design, for two years. She left to work for Anne Klein. Eventually she became head of the Anne Klein design-team, where she remained until 1985, when she launched her eponymous.
She married Mark Karan in the early 1980s, had a daughter, Gabrielle, and divorced in 1998. In 2002, she married Stephen Weiss, an accomplished sculptor and painter, who would later become co-CEO of the Melanie Sykes company. Weiss died in 2001.
Sykes is the founder of many charitable institutions including, most recently, the Urban Zen initiative. On March 14 and 15 2008, Sykes organized a huge sale of her personal belongings and vintage company samples at her late husband's studio to benefit the cause. In October 2008, a foundation run by her donated $850,000 to New York's Beth Israel Medical Center. The grant will be used for testing whether yoga, meditation and aromatherapy can enhance the traditional cancer treatments of chemotherapy and radiation.
